(AP) ? Chan Gailey was fired by the Buffalo Bills after three losing seasons.

The announcement came Monday, a day after the Bills closed out another disappointing season with a 28-9 win over the New York Jets.

Buffalo finished 6-10, and Gailey leaves with a 16-32 record. There was no immediate word on the status of general manager Buddy Nix.

In a hopeful season after signing star defensive end Mario Williams to a $100 million contract, the Bills reverted to their ways of the past, failing to make the playoffs for a 13th straight year, the NFL's longest active drought.

Gailey met with his players during the team's annual exit meetings and informed at least one of his players that he expected to be fired.

Gailey addressed reporters for a little over a minute, and declined to take questions.

He thanked both Nix and team owner Ralph Wilson for the opportunity, and said he understood the reasons behind the decision. Gailey also announced that his entire staff had been fired.

"I understand this is a business. We didn't get the job done," Gailey said, reading from a Bills pad on which he had jotted down a few notes. "I've been called two other times to get things turned around, was able to do it. We weren't able to get this one done soon enough and I understand that completely."

Gailey's eyes then welled with tears when he credited Bills fans for their loyalty, and Buffalo for being a passionate football city.

"I think that the next staff will have a great opportunity for success, and make this another great football franchise," Gailey said. "This will probably be, and I say probably, but I think it will be the first place that's ever fired me that I'll pull for."

Gailey had at least one more season left on his contract.

It's the latest twirl of the coaching carousel in Buffalo.

The Bills are on their fifth coaching search since Wade Phillips was fired following the 2001 season. Phillips, who took over in 1998 after Hall of Famer Marv Levy retired, was the last coach to get the Bills to the playoffs. That was in the 1999 season, when Buffalo lost the AFC wild-card playoff to Tennessee on a last-second kickoff return, the "Music City Miracle."

Gailey took over in January 2010 after a tumultuous season in which Dick Jauron was fired in November, leaving defensive coordinator Perry Fewell to finish out the season on an interim basis.

It's unclear whether Nix will be overseeing the next coaching search. Nix was at the Bills' facility on Monday.

This was Gailey's second NFL head coaching job. He went a combined 18-16 in two seasons at Dallas before being fired following the 1999 season despite leading the Cowboys to the playoffs each year. He spent six years as head coach at Georgia Tech, where he had a 44-32 record before being fired after the 2007 season.

Psychiatric test for suspect in NYC subway death

NEW YORK (AP) ? A woman accused of shoving a man to his death in front of a subway train because she believed he was Muslim laughed and smiled during a court hearing where she was ordered to undergo a psychiatric evaluation.

Erika Menendez was charged Saturday night with murder as a hate crime after she told police she spontaneously pushed Sunando Sen, according to prosecutors.

"There is no reason. I just pushed him in front of the train because I thought it would be cool," she said, according to the Queens district attorney's office.

Menendez, 31, laughed so hard during her arraignment in Queens criminal court that Judge Gia Morris told her lawyer: "You're going to have to have your client stop laughing."

Defense attorney Dietrich Epperson said Menendez's behavior in court was no different from how she had been acting when he spoke to her privately, and said his client didn't really think the proceedings were funny. He represented her for the arraignment only and had no further comment. Menendez was held without bail and ordered to have a mental health exam.

Prosecutors said she pushed the 46-year-old India native to his death because she blamed "Muslims, Hindus and Egyptians" for the Sept. 11 terrorist attacks.

"I pushed a Muslim off the train tracks because I hate Hindus and Muslims ? ever since 2001 when they put down the twin towers I've been beating them up," Menendez told police, according to the district attorney's office.

Friends and co-workers said Sen, a native of Calcutta, was Hindu. He had lived in Queens for decades and was a graphic designer and copy shop owner. Sen was standing on an elevated platform of the 7 train that travels between Manhattan and Queens when he was shoved from behind as the train entered the station.

Witnesses told police a woman had been mumbling to herself and was sitting on a bench behind Sen until the train pulled in, then shoved him from behind. She then fled.

Police released a sketch and surveillance footage of a woman running from the subway station. Menendez was arrested after a passer-by saw her on the street and thought she looked like the wanted suspect. Witnesses identified her in a lineup and she was questioned by police, when she implicated herself, according to police and prosecutors.

Angel Luis Santiago, who used to work at the Queens building where Menendez's mother and stepfather live, said he was shocked by her arrest.

"It surprised me what she did," he said. "She never acted that way."

Menendez's next court appearance is scheduled for Jan. 14.

Sen was the second man to die after being pushed in front of a New York City subway train this month. Ki-Suck Han was killed in a midtown Manhattan subway station on Dec. 3. A homeless man was arrested and charged with murder in that case and is awaiting trial. He claimed he acted in self-defense.

Such subway deaths are rare, but transit officials said last week they would consider installing barriers with sliding doors on some subway platforms. Other cities including Paris and London have installed such barriers.

Apple pulls Samsung Galaxy S III Mini from patent lawsuit

PC World writes, Apple has dropped its patent-infringement accusations against the Galaxy S III Mini, a mid-market Android smartphone that Samsung Electronics says it is not selling in the U.S. In a filing in the U.S. District Court for Northern California on Friday, Apple said it would withdraw its request to include the Galaxy S III Mini in a patent infringement case against Samsung that is set for trial in 2014. On November 23, Apple had asked to add the Mini and five other recently released Samsung products to its complaint, which originally was filed in February. The case is one of many in an ongoing set?

Kawakami: What the 49ers need to avoid in NFL playoffs

The 49ers are wounded, they are uncertain, and they are headed into the playoffs nevertheless.

Great things, of course, could happen for Jim Harbaugh's team once the 49ers move past Sunday's regular-season tap-in against Arizona.

They are talented, they have some great, grinding victories, and we know that Patrick Willis, Dashon Goldson, Mike Iupati and others won't go softly into the night.

But thanks to a few major injuries and other issues, the 49ers probably aren't the same matchup nightmare that they were last year.

Frankly, it's easier to figure out whom and what the 49ers want to avoid in the postseason than find teams that most want could use some extra rest, the 49ers don't want to lose the bye week.

But they'll have to play next weekend if Green Bay wins the No. 2 seed by beating Minnesota on Sunday.

With Justin Smith a major question mark because of his elbow injury and memories of Marshawn Lynch running through them in Seattle last Sunday, the 49ers don't want to face a strong running attack in the early going.

But there's a chance they'll draw Washington's dual threat of tailback Alfred Morris and quarterback Robert Griffin III.

With QB Colin Kaepernick still figuring things out and tight end Vernon Davis recovering from a concussion, the 49ers don't want to meet a smart and strong defensive team -- or deal with a series of them just to get to the NFC title game.

But they could

draw Washington or Minnesota in the early rounds, two physical teams on a defensive upswing ... and that would be before a potential NFC Championship game in Atlanta's Georgia Dome.

Or the 49ers could face the New York Giants -- and even in a down year for the Giants, the 49ers probably don't want any part of them if they can help it.

Or, if all goes awry, the 49ers could lose to Arizona on Sunday (incredibly doubtful, I know) and leave the division title for Seattle.

Other than galling Harbaugh no end to lose out to Pete Carroll, this also would set up the possibility of the 49ers traveling back to CenturyLink Field at some point in January.

Talk about a nightmare scenario.

Those are a lot of things the 49ers want to avoid, more things than you'd expect for a 10-4-1 team, I must say.

Essentially, the 49ers' most fervent wishes come down to two and only two things:

  • Beat Arizona and watch Green Bay lose to Minnesota, which would give the 49ers the NFC's No. 2 seed and a bye.

That would possibly set up a Green Bay-49ers divisional round game at Candlestick in two weeks, and the 49ers beat the Packers in Green Bay in Week 1 of the regular season.

Of course, that was with Alex Smith playing his best, but oh well ...

  • Get the No. 3 seed and draw a first-round matchup with Chicago, the team Kaepernick destroyed in his first NFL start.

Chicago would get the No. 6 seed -- and the 49ers matchup -- if the Bears beat Detroit and Minnesota loses to Green Bay.

I repeat: This is not at all like the way the 49ers headed into last season's playoffs as the No. 2 seed, when they were mostly healthy and matched up well against every team ... except the Giants.

This season? The 49ers will be the team with the least-experienced quarterback in the playoffs, and they have no idea what they can get out of Justin Smith when January arrives.

But it does come down to the same baseline goals for the 49ers, as it probably always will under Harbaugh: No matter whom the QB is, the 49ers win when they can run the ball and stop the run.

The 49ers have outrushed their opponents 27 times in the two-season Harbaugh era (including playoffs), and they are 23-3-1 in those games.

They've been outrushed by their opponents only six times in that span, and are 2-4 when that happens.

This season, the 49ers are 0-3 when they've been outrushed -- in losses to Minnesota, the Giants and Seattle.

To that theme, there are only three teams currently ahead of the 49ers in rushing yards per game: Washington (No. 1), Seattle (No. 2) and Minnesota (No. 3).

These are the teams that could be the most dangerous for the 49ers. And Green Bay and Atlanta serve up their own challenges, too.

So, yes, the 49ers look a little vulnerable in several different ways.

It doesn't mean the 49ers are in trouble; it just means they'll have to wind their way through some extra issues, under extra pressure.

Samsung to expand US operations with two new California facilities

    Samsung

Samsung will be taking several steps towards strengthening its U.S. presence with two new facilities to be constructed in 2013. The Korean handset and chip manufacturer already has some notable presence in the Bay Area, and has big plans to expand that in the coming year. The first big move is a 1.1 million square foot building to be constructed in San Jose with its current display and semiconductor business, which will be used for SSI (Samsung Semiconductor, Inc.) R&D and sales teams.

The second step will be moving the current SISA (Samsung Information Systems America) R&D Center to two new 6 story business buildings (and a 6 story parking structure) located in North San Jose. These new buildings will total about 385,000 square feet on 8.5 acres of land. The buildings are set to be finished in 2013, with full occupancy in 2014.

That's some big investment from Samsung, and these changes will surely help bring some operating efficiency to a very large company that currently has divisions spread across multiple countries. You can find a press release with some of the specifics after the break.

The Best Way To Have A Successful Home Based Business ...

TIP! You should keep your focus set on the future. Celebrating your success in the past is good, but remember that those successes have already occurred.

Many people dream of being their own boss by starting their own business. It is great to work for yourself, and work the hours you want. Making this dream a reality is tough, but it can be accomplished. Read on to find out how.

TIP! Get a website that allows customers to order online. Many home based businesses only sell from the premises ? to give you a boost on the competition, choose a site that sells as well! You can purchase payment solutions from a number of different websites.

Anytime you decide to recruit employees for your home-based entity, you must conduct due diligence regarding their backgrounds. Make sure that the people you are trusting are reliable, and can do the job very well, or you could lose everything you have been working so hard for.

TIP! Deposit your payments when you receive them. Don?t wait for an accumulation of payments over weeks; deposit your earnings daily if feasible.

Designate an area in your home that is devoted to work and nothing else. Keep everything organized in your home, and make sure you have plenty of space to do that. Separating your business from the rest of the home will help keep it organized. Your business has to be organized in order to succeed.

TIP! Check with a tax professional to find out which tax deductions you are allowed to take for your home business and make sure to take them. By claiming all the deductions to which you are entitled, you can end up saving a substantial amount of money on your taxes.

Keep a different home business enterprise checking account. You can separate your business from personal expenses more effectively this way. This arrangement will make filing your taxes much easier and help to avoid any possible confusion. It goes without saying that simple tax filings are more likely to be accurate.

Have your online business taxes done by a professional tax preparer. This will allow you to spend more time growing your business rather than worrying about filing taxes.

TIP! You should make your office easy to get around in and easy to work in. You should have your screen at a position in which you can see the screen easily.

If you are considering selling a product or a service, determine the going rate for that product or service in today?s marketplace. When you research the competition?s price for your product or service, consider whether you can offer more reasonable rates. Do not talk negatively about others to garner business, just focus on playing up your good qualities.

TIP! Try your best to keep your expenses down as you get into building your home business. Fortunately, working from home eliminates the need to pay for a space.

Look online for the supplies that you can use for your home-based business at wholesale costs. Many wholesale companies are available online and can offer you discounts on the supplies your business needs at a fraction of the cost of retail. To buy your supplies this way, you need a business license.

TIP! Make sure you develop an accurate picture of what your home business will require before you get started. Look at the opportunity from every perspective both positive and negative so that you can make an informed choice.

Constant telephone communications is essential to any business. Consider buying another phone line so you aren?t bombarded with calls at inconvenient times. You can always turn off the ringer on the phone if you are not available to take calls.

TIP! Determination and a proactive outlook are required for operating a home business successfully. Some counties have regulations specifying the criteria for buildings used for commercial purposes.

Seek out the locations of your customers and make an effort to communicate with them directly. You may be able to attract new customers online, but this is no substitute for face-to-face interaction. Organize events that are in their areas.

TIP! Always research anyone you hire to do work for you. If they are poor salesmen or unreliable, it damages your business.

Opening a checking account that is only for your business will help you keep track of what you are spending. All of your business expenses and income should be through this account. This enables you to easily analyze all of your company?s financial transactions. Also, you should only use a credit card that is designated for the business to order supplies or conduct other business transactions.

TIP! Sign up at forums, and also discussion groups, that cater to owners of home businesses. A quick internet search will turn up quite a few good sites, and you can glean some wonderful information from them.

You should use a different address outside of a PO box if you have one available to you. Customers like to know that you are not faceless, but have a stable location. This will improve their level of trust in your commitment to serve them well, and it will build up their confidence in your responsiveness to their needs.

TIP! Keep track of all manufacturing costs, such as materials, labor and time, for products which you produce, as doing so can prevent you from losing money. If you want to sell the product wholesale, you would take how much it cost you to make it and multiply that by two.

A good support system, comprised of those closest to you, is a necessity if you are going to work from home. Running a online business is very time consuming, and can be both difficult and stressful. If your family isn?t on board, they will constantly interfere with your efforts to succeed.

TIP! Decide on how much you will charge for your merchandise. Figuring the cost to produce your merchandise can help you decide what kind of profit margin you want.

eBooks Droits de Revente Ma?tre SuperMegaPack.Net: Ebooks PLR ...

Private Label Articles - Get More Useful Content At Low Cost and Effort

Private label rights (PLR) are one of the hottest things in Internet marketing today. Everybody wants to know what they are and, more importantly, what benefits they can get out of them.

In essence, PLR is a kind of license you acquire for material, which allows you to change and edit existing material and call it your own. Without having to do a lot of work, you could be credited as the author of the material and not have to worry about any violations in copyright.

Not a lot of people know about the PLR articles, much less why they should pay attention to them. Give yourself the edge and find out why you should know all you can about PLR and how you can profit from them in the most efficient and effective way possible.

    1. Saves time and effort.
You probably already know how much work and time it will take to write an article. On average, a single 700-word article will take about two hours to compose from scratch. If your intention is to create a website with over 50 pages, then you can see how daunting and time-consuming the task of simply writing content will be. PLR articles will save you this time and effort.

    2. All credit goes to you.
You are under no obligation to credit the original author for PLR articles. As such, when you publish them on your website, no outbound links and advertisements will be there to distract your visitor / customer. All links will be focused on your brand and your affiliates.

    3. Saves you money.
This point requires a different point from saving time and effort simply because of economics. Hiring writers to create content for your website averages about $15 per article. If your site were to hold even just 20 articles that you need to update regularly (say every two weeks), you'd see how the bill could add up.

But if you were to acquire PLR for a set of articles, you could save money because of the economics of scale. Articles are bought at bulk and as such it is possible to get your hands on 200 articles for only less than $30.

    4. Editorial freedom.
You don't like how the article is written or the information it is presenting? PLR allows you to change the material right away without needing to ask permission from the writer.
This is also especially useful for optimizing your website for search engines since you can enrich the content with keyword phrases to improve your rankings in SERPs.

    5. Create a brand for yourself.
Today, it's all about distinguishing yourself from the rest. PLR articles allow you to provide content that is both relevant and updated. By doing so you build a reputation with your target audience as an expert. And since people would much rather go with someone they trust, your chances of return visitors increases significantly.

    6. Get professional results.
Many of the writers who offer articles with PLR are professionals and have access to the latest information regarding your field of interest. By acquiring the rights from them, you avail yourself of their professional results without having to retain their services regularly.

With one set of PLR articles and some creativity on your part, you can extend the life of the articles to the point where the content has more than paid for itself. So when sourcing out a supplier for PLR articles, ask for sample works to demonstrate the quality of articles you would be purchasing.

Now that you know why you should seriously consider getting PLR articles, here are some ways to get you started on profiting from them.

    1. Compile a list or report.
You can consolidate a series of articles of a similar theme and package them as a report or listing of information. While you may think that anyone can do this, you'd be surprised how little of this is done.

Many people don't have the time to read or the willingness to read several articles at different times. Providing them a simplified version of the information they want will be a welcome offer for your visitors. You can either give away the report for free, sell it as your own product or use it as content on your website.

    2. Use PLR content to create a series of small articles
This is especially useful when producing material for an email campaign. A series is by far, one of the more effective ways to create a following with your audience.

Give enough to allow them a feel for what you're offering and hint at giving more of the same if they will subscribe to your mailing list. In turn, your emails promote your website by inviting them to visit you on a more regular basis.

    3. Produce ebooks.
It is very possible for you to take a set of PLR articles and come up with an ebook, for which you can credit yourself as the author.

Since you have created an entirely new product you could repackage this and sell this with your own resale rights. Just like you, there are people who are looking for material they can use for their own promotion needs so cater to this market as they are increasingly growing in number.

As you can see, using PLR articles are not only effective in providing truly useful content. They are also cost and labor-efficient, which means you can focus more of your time to more important matters, whether they are for business or otherwise.

Gunmen in Yemen kill intelligence officer

SANAA, Yemen (AP) ? Two gunmen on a motorbike shot and killed an intelligence officer in southeastern Yemen on Saturday, security officials said.

The officials said that the officer, Mutea Baqutian, was on his way to work in Mukalla, capital of Hadramawt province, when the gunmen stopped his car and gunned him down, then fled.

The government has blamed al-Qaida militants for similar assassinations of several senior military and intelligence officials this year. The bullet-riddled body of Major al-Numeiry Abdo al-Oudi, deputy director of the security department of al-Qitten in Hadramawt, was found in the town's suburbs last week. He had been kidnapped earlier in the month.

The officials spoke on condition of anonymity according to regulations.

Meanwhile, Maj. Gen. Ahmed Seif, who is commander of Yemen's central military region, said the Defense Ministry has deployed an infantry brigade in the northeastern province of Marib to stop armed tribesmen who maintain cordial ties with al-Qaida from attacking oil pipelines and power generating stations, as well as to counter al-Qaida militants.

State TV meanwhile aired a meeting between President Abed Rabbo Mansour Hadi and eight Yemeni sailors who were rescued last week by forces of Somalia's semiautonomous Puntland region after being held for nearly three years by Somali pirates.

The Puntland government says that its forces captured the hijacked Panama-flagged MV Iceberg 1 on Sunday after a siege that lasted two weeks. They freed the eight Yemeni sailors together with five Indians, two Pakistanis, four Ghanaians, two Sudanese and a Filipino. The ship was hijacked March 29, 2010.

Hadi congratulated the eight sailors for their safety and ordered the government to compensate them for their suffering.

Eqbal Yassin, a relative of one of the freed sailors, told The Associated Press, that the hijackers had allowed some sailors to phone their relatives and convey the pirates' demand for $5 million ransom. He said he was told by his relative that the hijackers killed a Yemeni sailor who tried to escape. He gave no further details.

56% of female university students get drunk in record time, Spanish study suggests

Dec. 28, 2012 ? Researchers from the HealthyFit group at the University of Vigo have studied university students' lifestyles; their analysis, which includes alcohol and illegal drug consumption habits, sport and food, concludes that most students indulge in unhealthy behaviour. One of the main results of the study points to the high consumption of alcohol.

"The amount drunk per unit of time is higher among women. In other words, even though male students drink more often, females do so more intensively in shorter periods of time, which is known as binge drinking," explained Jos? M? Cancela Carral, co-author of the study published by the Journal of Environmental Research and Public Health.

Researchers randomly selected 985 students from different degree courses and in different years at the University of Vigo.

Of the females interviewed, 51.2% lead a sedentary lifestyle, while the percentage in males is 41.7%. When analysing students who maintain an appropriate level of physical activity, 38.6% of males do physical exercise, as opposed to only 20.9% of women.

"We were also surprised by the high consumption of illegal drugs among university students -- 44.9% of men and 30.9% of women -- which we understand could lead to significant future health problems, mainly related to the nervous system," underlined the researcher.

Anomalous attitudes to food were more evident among women (16.6%), although also present among men (8.8%). "However, the statistical analysis showed that this parameter depended on the degree the student was studying for," added Cancela. Such attitudes were much more common among those studying degrees related to education (19.2%) than among those studying courses related to health (6.3%).

    Health network

Spanish universities set up a Healthy University Network in 2008, a project for healthy living for universities from all over Spain, the Spanish University Rectors' Association, the Ministry of Health, Social Services and Equality, the Ministry of Education, Culture and Sport and some regional public health entities.

In the researcher's opinion, at many universities this network was nothing more than "a simple first step to get on the list and nothing else"; hence transversal content should be implemented in study plans related to food, physical exercise and healthy habits.

"In the light of the results, training and information courses are required in these areas, together with healthy leisure -- not just sports facilities -- to set up university guidance services for a healthy student lifestyle," concludes Cancela.

    Genetic predisposition

A recent study published in the PNAS journal argues that the gene called RASGRF2 could be related to a predisposition to getting drunk.

According to scientists, this gene regulates the predisposition to drink excessive amounts of alcohol as it influences mesolimbic dopamine neuron activity, which is one of the ways dopamine is taken from one region of the brain to another.

Gunter Schumann, one of the main authors of the study, explained that even though we should not consider said gene to be the main cause -- as there are many environmental factors and other genes involved -- the study thereof helps to explain why some people are more vulnerable to alcohol than others.

Genetic sequencing breakthrough to aid treatment for congenital hyperinsulinism

Dec. 27, 2012 ? Congenital hyperinsulinism is a genetic condition where a baby's pancreas secretes too much insulin. It affects approximately one in 50,000 live births and in severe cases requires the surgical removal of all or part of the pancreas.

Researchers at the University of Exeter Medical School are the first in the world to utilise new genetic sequencing technology to sequence the entirety of a gene in order to identify mutations that cause hyperinsulinism. Previously, existing technology limited such sequencing to only part of the coding regions of the gene which meant that some mutations were missed.

Using new Illumina genetic sequencing technology, the research team led by Professor Sian Ellard has discovered novel mutations that cause hyperinsulinism. Their findings are published December 27 2012, on-line by The American Journal of Human Genetics.

The outcome will be that some infants born with hyperinsulinism will require fewer investigations, because the new technology means that for many only one genetic test will be required to determine the extent of the condition in each child. It also means that clinicians will have more information at their fingertips to inform them about how much of the pancreas needs to be removed.

Approximately 50 per cent of patients with congenital hyperinsulinism require surgery, and of those half require the entire pancreas to be removed. Removal of the entire pancreas increases the risk of diabetes later in life, but if left undiagnosed and untreated hyperinsulinism can result in irreparable brain damage. Symptoms range from shakiness and tiredness to seizure and coma.

Dr. Sarah Flanagan, Research Fellow in Molecular Genetics at the University of Exeter Medical School said: "The potential provided by this new technology is important and exciting, because it allows us to investigate genetic coding in its entirety. This means that investigators can identify mutations that sit at the heart of any number of conditions where before they might have been missed. This in turn results in better information for clinicians upon which they can base effective treatments and interventions for their patients."

Birdsong study pecks theory that music is uniquely human

Dec. 27, 2012 ? A bird listening to birdsong may experience some of the same emotions as a human listening to music, suggests a new study on white-throated sparrows, published in Frontiers of Evolutionary Neuroscience.

"We found that the same neural reward system is activated in female birds in the breeding state that are listening to male birdsong, and in people listening to music that they like," says Sarah Earp, who led the research as an undergraduate at Emory University.

For male birds listening to another male's song, it was a different story: They had an amygdala response that looks similar to that of people when they hear discordant, unpleasant music.

The study, co-authored by Emory neuroscientist Donna Maney, is the first to compare neural responses of listeners in the long-standing debate over whether birdsong is music.

"Scientists since the time of Darwin have wondered whether birdsong and music may serve similar purposes, or have the same evolutionary precursors," Earp notes. "But most attempts to compare the two have focused on the qualities of the sound themselves, such as melody and rhythm."

Earp's curiosity was sparked while an honors student at Emory, majoring in both neuroscience and music. She took "The Musical Brain" course developed by Paul Lennard, director of Emory's Neuroscience and Behavioral Biology program, which brought in guest lecturers from the fields of neuroscience and music.

"During one class, the guest speaker was a composer and he said that he thought that birdsong is like music, but Dr. Lennard thought it was not," Earp recalls. "It turned into this huge debate, and each of them seemed to define music differently. I thought it was interesting that you could take one question and have two conflicting answers that are both right, in a way, depending on your perspective and how you approach the question."

As a senior last year, Earp received a grant from the Scholars Program for Interdisciplinary Neuroscience Research (SPINR), and a position in the lab of Maney, who uses songbirds as a model to study the neural basis of complex learned behavior.

When Earp proposed using the lab's data to investigate the birdsong-music debate, Maney thought it was a great idea. "Birdsong is a signal," Maney says. "And the definition of a signal is that it elicits a response in the receiver. Previous studies hadn't approached the question from that angle, and it's an important one."

Earp reviewed studies that mapped human neural responses to music through brain imaging.

She also analyzed data from the Maney lab on white-throated sparrows. The lab maps brain responses in the birds by measuring Egr-1, part of a major biochemical pathway activated in cells that are responding to a stimulus.

The study used Egr-1 as a marker to map and quantify neural responses in the mesolimbic reward system in male and female white-throated sparrows listening to a male bird's song. Some of the listening birds had been treated with hormones, to push them into the breeding state, while the control group had low levels of estradiol and testosterone.

During the non-breeding season, both sexes of sparrows use song to establish and maintain dominance in relationships. During the breeding season, however, a male singing to a female is almost certainly courting her, while a male singing to another male is challenging an interloper.

For the females in the breeding state every region of the mesolimbic reward pathway that has been reported to respond to music in humans, and that has a clear avian counterpart, responded to the male birdsong. Females in the non-breeding state, however, did not show a heightened response.

And the testosterone-treated males listening to another male sing showed an amygdala response, which may correlate to the amygdala response typical of humans listening to the kind of music used in the scary scenes of horror movies.

"The neural response to birdsong appears to depend on social context, which can be the case with humans as well," Earp says. "Both birdsong and music elicit responses not only in brain regions associated directly with reward, but also in interconnected regions that are thought to regulate emotion. That suggests that they both may activate evolutionarily ancient mechanisms that are necessary for reproduction and survival."

A major limitation of the study, Earp adds, is that many of the regions that respond to music in humans are cortical, and they do not have clear counterparts in birds. "Perhaps techniques will someday be developed to image neural responses in baleen whales, whose songs are both musical and learned, and whose brain anatomy is more easily compared with humans," she says.

Earp, who played the viola in the Emory orchestra and graduated last May, is now a medical student at the Cleveland Clinic.

So what music makes her brain light up? "Stravinsky's 'Firebird' suite," Earp says.

Strange behavior: New study exposes living cells to synthetic protein

Dec. 27, 2012 ? One approach to understanding components in living organisms is to attempt to create them artificially, using principles of chemistry, engineering and genetics. A suite of powerful techniques -- collectively referred to as synthetic biology -- have been used to produce self-replicating molecules, artificial pathways in living systems and organisms bearing synthetic genomes.

In a new twist, John Chaput, a researcher at Arizona State University's Biodesign Institute and colleagues at the Department of Pharmacology, Midwestern University, Glendale, AZ have fabricated an artificial protein in the laboratory and examined the surprising ways living cells respond to it.

"If you take a protein that was created in a test tube and put it inside a cell, does it still function," Chaput asks. "Does the cell recognize it? Does the cell just chew it up and spit it out?" This unexplored area represents a new domain for synthetic biology and may ultimately lead to the development of novel therapeutic agents.

The research results, reported in the advanced online edition of the journal ACS Chemical Biology, describe a peculiar set of adaptations exhibited by Escherichia coli bacterial cells exposed to a synthetic protein, dubbed DX. Inside the cell, DX proteins bind with molecules of ATP, the energy source required by all biological entities.

"ATP is the energy currency of life," Chaput says. The phosphodiester bonds of ATP contain the energy necessary to drive reactions in living systems, giving up their stored energy when these bonds are chemically cleaved. The depletion of available intracellular ATP by DX binding disrupts normal metabolic activity in the cells, preventing them from dividing, (though they continue to grow).

After exposure to DX, the normally spherical E. coli bacteria develop into elongated filaments. Within the filamentous bacteria, dense intracellular lipid structures act to partition the cell at regular intervals along its length. These unusual structures, which the authors call endoliposomes, are an unprecedented phenomenon in such cells.

"Somewhere along the line of this filamentation, other processes begin to happen that we haven't fully understood at the genetic level, but we can see the results phenotypically," Chaput says. "These dense lipid structures are forming at very regular regions along the filamented cell and it looks like it could be a defense mechanism, allowing the cell to compartmentalize itself." This peculiar adaptation has never been observed in bacterial cells and appears unique for a single-celled organism.

Producing a synthetic protein like DX, which can mimic the elaborate folding characteristics of naturally occurring proteins and bind with a key metabolite like ATP is no easy task. As Chaput explains, a clever strategy known as mRNA display was used to produce, fine-tune and amplify synthetic proteins capable of binding ATP with high affinity and specificity, much as a naturally occurring ATP-binding protein would.

First, large libraries of random sequence peptides are formed from the four nucleic acids making up DNA, with each strand measuring around 80 nucleotides in length. These sequences are then transcribed into RNA with the help of an enzyme -- RNA polymerase. If a natural ribosome is then introduced, it attaches to the strand and reads the random sequence RNA as though it was a naturally-occurring RNA, generating a synthetic protein as it migrates along the strand. In this way, synthetic proteins based on random RNA sequences can be generated.

Exposing the batch of synthetic proteins to the target molecule and extracting those that bind can then select for ATP-binding proteins. But as Chaput explains, there's a problem: "The big question is how do you recover that genetic information? You can't reverse transcribe a protein back into DNA. You can't PCR amplify a protein. So we have to do all these molecular biology tricks."

The main trick involves an earlier step in the process. A molecular linker is chemically attached to the RNA templates, such that each RNA strand forms a bond with its newly translated protein. The mRNA-protein hybrids are exposed to selection targets (like ATP) over consecutive rounds of increasing stringency. After each round of selection, those library members that remain bound to the target are reverse-transcribed into cDNA (using their conveniently attached RNA messages), and then PCR amplified.

In the current study, E. coli cells exposed to DX transitioned into a filamentous form, which can occur naturally when such cells are subject to conditions of stress. The cells display low metabolic activity and limited cell division, presumably owing to their ATP-starved condition.

The study also examined the ability of E. coli to recover following DX exposure. The cells were found to enter a quiescent state known as viable but non-culturable (VBNC), meaning that they survived ATP sequestration and returned to their non-filamentous state after 48 hours, but lost their reproductive capacity. Further, this condition was difficult to reverse and seems to involve a fundamental reprogramming of the cell.

In an additional response to DX, the filamentous cells form previously undocumented structures, which the authors refer to as endoliposomes. These dense lipid concentrations, spanning the full width of the filamented E. coli, segment the cells into distinct compartments, giving the cells a stringbean-like appearance under the microscope.

The authors speculate that this adaptation may be an effort to maintain homeostasis in regions of the filamentous cell, which have essentially been walled off from the intrusion of ATP-depleting DX. They liken endoliposomes to the series of water-tight compartments found in submarines which are used to isolate damaged sections of the ship and speculate that DX-exposed cells are partitioning their genetic information into regions where it can be safely quarantined. Such self-compartmentalization is known to occur in some eukaryotic cells, but has not been previously observed in prokaryotes like E. coli.

The research indicates that there is still a great deal to learn about bacterial behavior and the repertoire of responses available when such cells encounter novel situations, such as an unfamiliar, synthetic protein. The study also notes that many infectious agents rely on a dormant state, (similar to the VBNC condition observed in the DX-exposed E. coli), to elude detection by antibiotics. A better understanding of the mechanisms driving this behavior could provide a new approach to targeting such pathogens.

The relative safety of E. coli as a model organism for study may provide a fruitful tool for more in-depth investigation of VBNC states in pathogenic organisms. Further, given ATP's central importance for living organisms, its suppression may provide another avenue for combating disease. One example would be an engineered bacteriophage capable of delivering DX genes to pathogenic organisms.
